It is now well established that beyond their essential role as components of protein, amino acids, possibly acting via nutrient transporters/receptors (“transceptors”) or intracellular sensors, have the capability to initiate and/or regulate crucial cell signalling pathways that control a diverse array of cellular processes including, for example, gene expression, autophagy, lysosomal biogenesis, and lipid and protein synthesis that ultimately influence cell growth and proliferation. Regulation of many of these cellular processes is critically dependent on the activity of the mTOR complex 1 (mTORC1) signalling complex, which is responsive not only to hormonal cues but to changes in amino acid availability. While the past decade has seen significant developments in our understanding of the sensing and signalling mechanisms by which amino acids influence mTORC1 activity the editorial team recognise that future advances in this research area could help inform the development of strategies that utilize amino acids as nutraceuticals to counter certain pathologies such as sarcopenia. Alternatively, studies in this area may help identify novel amino acid sensors as therapeutic targets for suppressing mTORC1, which may help in the treatment of certain cancers and neurodegenerative conditions.
The editors of AJP - E&M invite submission of original research articles that provide novel mechanistic insight of how particular amino acids or their metabolic derivatives are sensed by cells, how they may influence activation and cross-talk between signaling pathways regulating key aspects of cell and tissue function and whether use of amino acids or specific pharmacological derivatives can confer therapeutic benefits for disease treatment. We will consider both original research articles and review articles.
